Orforglipron weight-loss results: understanding ATTAIN-1

Research results are easier to use when you know who took part, how long treatment lasted and what an average can tell you.

What did the trial study?

ATTAIN-1 enrolled 3,127 adults with obesity without diabetes. Participants received daily investigational orforglipron or placebo for 72 weeks, with diet and activity support. The study tested investigational doses of 6, 12 and 36 mg. ATTAIN-1: orforglipron in adults with obesity (NEJM, 2025) ↗

Average changes at 72 weeks

ATTAIN-1 treatment-regimen analysis
Study groupAverage body-weight change
Orforglipron 6 mg−7.5%
Orforglipron 12 mg−8.4%
Orforglipron 36 mg−11.2%
Placebo−2.1%

This analysis accounts for treatment discontinuation. Digestive adverse events were the most common and were usually mild or moderate. Treatment discontinuation because of adverse events occurred in 5.3–10.3% of orforglipron groups, compared with 2.7% of the placebo group.

How should a patient read the numbers?

A percentage describes change relative to starting weight. As an arithmetic example, an 11.2% change from 100 kg is 11.2 kg. That calculation explains the unit; it does not predict what a person starting at 100 kg will lose. A group average combines people with different responses.

The 72-week endpoint also matters. It is not a one-month result, and it is not a reason to promise a particular amount of weight loss by an appointment date. Ask your clinician how progress will be assessed during your own follow-up rather than choosing a target from an online headline.

Trial doses are not a tablet-conversion guide

Do not match the investigational milligram numbers above directly to marketed tablet strengths. Formulation and prescribed dosing instructions matter. This trial did not test Orfoxen-branded tablets, so it does not establish Orfoxen-specific outcomes or equivalence to another brand.

A useful consultation question is: “Which evidence applies to my situation, and what do we know about the exact product being supplied?” Also ask what the plan would be if tolerability prevents the intended routine.

Keep the evidence and the practical plan together

Save the clinical evidence guide, your prescribed instructions and the current price page. They answer different questions: research context, your own plan and the practical cost. If you also have diabetes, ask which studies and treatment goals are relevant to that diagnosis.

مریضوں کے لیے اردو خلاصہ
